Dornseiff lehrte als Professor für klassische Philologie 1925-26 in Berlin, 1926-46 in Greifswald, 1946-48 in Erlangen sowie ab 1948 in Leipzig. In Leipzig war er außerdem Direktor des Philologischen Instituts.<br />

Seine Übersetzung und Kommentierung des griechischen Dichters Pindar gilt als wegweisend. Einer breiteren Öffentlichkeit wurde Dornseiff jedoch vor allem durch sein 1934 erstmals veröffentlichtes Werk ''Der deutsche Wortschatz nach Sachgruppen'' bekannt. Anders als in klassischen [[Semasiologie|semasiologischen]] [[Wörterbuch|Wörterbüchern]] ordnete er darin den deutschen Wortschatz nicht alphabetisch, sondern nach Sachgruppen. Dornseiffs Wortschatz nach Sachgruppen ist bis heute das bedeutendste [[Onomasiologie|onomasiologische]] Wörterbuch der deutschen Sprache.<br />
