Isotopien werden vor allem auf der semantischen Ebene untersucht (Greimas 1971) und bezeichnen dann das wiederholte Auftreten semantischer Merkmale in lexikalischen Einheiten eines [[Text]]es.<br />

Durch Semrekurrenz entstehen Isotopieketten. Ein [[Text]] ist semantisch (auch) ein Gefüge von Isotopieebenen. ''Lexeme, die in einem Text über ein gemeinsames, rekurrent-dominantes Merkmal verknüpft sind, konstituieren eine Isotopieebene. Eine Isotopieebene kann immer dann angenommen werden, wenn wenigstens zwei Lexeme diese Voraussetzung erfüllen'' (Kallmeyer u. a. 1974: 149).<br />

Rastier (1974) stellt den Isotopien auf semantischer Ebene, den ''Inhaltsisotopien'' (159), Isotopien auf anderen Ebenen gegenüber, z. B. auf der phonologischen Ebene (durch Alliteration oder Reim) (152). Rastier, der Isotopie als ''fundamentalen Begriff'' der [[Textanalyse]] ansieht, unterscheidet sememische (oder horizontale) Isotopien (160 ff) von metaphorischen (oder vertikalen) Isotopien (166 ff).<br />

Agricola hat bestimmte Verfahren entwickelt, um über Isotopieketten das [[Textthema]] erschließen zu können (Agricola 1983: 230 ff).<br />

Die Dominanz bestimmter Arten von Isotopieketten ist auch textsortenabhängig; so sind prototypische Elemente dominanter Isotopien in narrativen Texten z. B. Bezeichnungen der Handlungsträger.<br />
